IMO E.

Between D/E, E is better as discourage X from doing Y is the right idiom.
Getting defeated is just a temporary notion, giving it up is what makes it permanent.

Reactions from <a> and <b> did not discourage <c> from exhibiting...

(A) did not discourage the Primitivist painter
Henri Rousseau to exhibit repeatedly,
despite him completely lacking formal
training and starting late - here even if this is right, the HIM should be HE, so this cannot be the right answer

(B) did not discourage the Primitivist painter
Henri Rousseau exhibiting repeatedly,
completely lacking formal training though
he was, and starting late - not a proper formation, awkward construction

(C) did not discourage the Primitivist painter
Henri Rousseau to exhibit repeatedly,
despite their complete lack of formal
training and late start - THEIR refers to <a> and <b>, wrong reference

(D) did not discourage the Primitivist painter
Henri Rousseau in exhibiting repeatedly,
despite his complete lack of formal
training and late start - HR (<c>) "IN" exhibiting...? wrong construction

(E) did not discourage the Primitivist painter
Henri Rousseau from exhibiting repeatedly,
despite his complete lack of formal
training and late start. - Right idiom, right construction...HR (<c>) FROM exhibiting....
