The comma after that is necessary.
For example:

Jane is as happy as, if not happier than, many of her friends.

if the comma is removed, it creates an error.

Jane is as happy as, if not happier than many of her friends.
as happy as breaks link. It is not correct

IMO: C is correct because "he will not publish his poetry" is right.

A-the sentence should be divided by comma to make it clear. ...if not more gifted than, many of his colleagues,...
B- ...he is extremely modests with his poetry remaining unpublished is wordy and akward.
C-chi ching! correct answer.
D-despite his being gifted.... it is just wrong.
E-Being as gifted as..... it is just so wrong.

I would immediately remove all the options with the word "unpublished" and then analyze the remaining two options and remove the one with despite.. I think it quicker!!
