One of the shirts that are lying on the couch is mine.

Or
In what way I will have to write in 'that clause', if I have to describe one ?

In what way I will have to write in 'that clause', if I have to describe all shirts ?

Here we are concerned with "only one" shirt hence usage of "is" is correct.

Maybe this: "One of the many things that I have to do is water the plants"

Maybe like this: "All the shirts that you see in the closet are mine"

"Of all the shirts that you see on the table, the red one is mine"
