Laos has a land area about the same as Great Britain but only 4 million in population, where many are members of hill tribes ensconced in the virtually inaccessible mountain valleys of the north.

My questions -

If option E read as -

comparable to that of Great Britain but a population of only 4 million , many of whom

Would it still be correct?

Would it be incorrect because population is a collective noun and hence can't be referred back by them

Population is a collective noun and singular e.g. Population of Australia is increasing.

'Who' / 'whom' is used for human beings so is correct.
