Basic question on Coordinate Geometry

I have a very basic question with regard to the below formula for slope:

y2 - y1
-------
x2 - x1

My question is how do you determine which one is y2 & y1?

Lets say (0,-3) & (4,-5). Is y2 - 5 or -3?

Lets take another example, (8,0) & (4,-5). Which one is y2 in this case?

Pls clarify.

Doesn't matter as long as you remain consistent with x and y
