Pradeep,
both use for comparison, here we don't need to be narrow down based on use of as\that.

A letter by Mark Twain, written in the same year as The Adventures of Huckleberry Finn were published, reveals that Twain provided financial assistance to one of the first Black students at Yale Law School.

(A) A letter by Mark Twain, written in the same year as The Adventures of Huckleberry Finn were published, -->INCORRECT: 'The Adventures of Huckleberry Finn" is a book, which is singular and hence require 'was' instead of 'were'

(B) A letter by Mark Twain, written in the same year of publication as The Adventures of Huckleberry Finn,-->INCORRECT: seems illogical comparison

(C) A letter by Mark Twain, written in the same year that The Adventures of Huckleberry Finn was published, -->CORRECT: seems correct

As the part after underlined word is 'reveals', we should have a singular subject, which D\E fails to provide

(A) A letter by Mark Twain, written in the same year as The Adventures of Huckleberry Finn were published,
[The Adventures of Huckleberry Finn - is the title of the book and hence SINGULAR and needs singular verb WERE is wrong here.]
(B) A letter by Mark Twain, written in the same year of publication as The Adventures of Huckleberry Finn,
[The sentence should say the letter was written in the same year in which the book was published . Some how the sentence formation not proper.]
(C) A letter by Mark Twain, written in the same year that The Adventures of Huckleberry Finn was published,
[THAT here refers to refres to the year in which the book was published .This is better sentence than B and the correct choice.]
(D) Mark Twain wrote a letter in the same year as he published The Adventures of Huckleberry Finn that
[THAT here refers to the book and changes the meaning. As if the book reveals some thing and not the letter. ]
(E) Mark Twain wrote a letter in the same year of publication as The Adventures of Huckleberry Finn that
[THAT here refers to the book and changes the meaning. As if the book reveals some thing and not the letter. ]

THAT can be used used as a pronoun .

Ex : That was a great shot .

THAT can also be used as adjective .
