Hi,
I have 5.5 years of experience. My current job demands more than 12 hours of work per day.Most of the time I am working on weekends also. I took GMAT last Oct and scored a miserable 640. With the amount of work pressure I don't think I will be able to concentrate and get a respectable score.
I am thinking to quit and study for 3 months , take GMAT and then search for another job.
Can this break have an adverse effect to my MBA application ?

Well, can you do it without having a "break in your resume"? Meaning talk to your employer and ask for a three month GMAT sabbatical?
something like that would be best.
